For me it's the first line in the series: "Space...it seems to go on and on forever...but then you get to the end and a giant monkey throws barrels at you."

Here's mine:

Leela: Don't you ever think ahead?

Fry: Hell No! If I ever stopped to think ahead, I wouldn't be emperor, and I would be here in the year 3000. It's just like the story about the grasshopper and the octopuss. All year long the grasshopper buried it's acorns for winter, while the octopuss mooched off his girlfriend and watched T.V. Then winter came, the grasshopper died. And the octopuss got all of his acorns, and he also got a racecar! Is this story getting through to you?

Fry: "God Help Us!"

It just sums up what an idiot he is. Also, the idea that they're on a robot populated planet; watching a film that's a reversal of 50s B-flicks; shows the odd humor and style of the whole series.

For me, it was the bit in the first ep. when Fry goes 'Cool! Like in Star Trek!' when he walks through the door and it slams in his face.

i believe the joke that defined futurama for me would have to be the "but Bender need brain... for... smart making" from How Hermes requisitioned his groove back.

in, I Roomate, when there in the escher room, the line "i dont think we want to pay for a demension we cant use".  that defines futurama for me, i dont know why.
