Well, it has been hinted that Leela may be something of a heavy drinker (started brewing her own at the orphanarium, loves the taste of a bold, Canadian beer early in the morning). To me that would just add to her pathos.
As for doing illegal drugs - I don't see it with her; she's way too concerned about keeping her body in fantatic physical condition (although I wonder if she has done steroids - the videogame suggests that she has).
Fry may have experiemented in the 20th century, but that would be as far as he has gone (but he does get drunk with Bender way too often).
Hermes definitely likes to get high - Jamacians have other interest and that's why they got held at the airport during the Olympics. And according tohis wife, he can't go anywhere without lighting something up.
Amy? I don't see it with her. Sure she's young and shallow, but she seems to be able to make her own fun without the aid of getting high.
The Professor. Sure, the old goat probably sniffs glue and burning Rhesus monkeys to get the day started.
Scruffy. You bet. He probably does some poppers while he's thumbing through his porno mags.

Amy doesn't even like coffee. (See "Put Your Head on My Shoulder".) So obviously she knew what "coffee" meant, especially in a back seat, or on Valentine's Day.
That said, there's no evidence of Amy doing mind-altering drugs other than alcohol; ditto for Leela. A lot of drinking goes on in _Futurama_, and not all of it by Bender and Fry.
That said again, who knows what kind of mind alteration is considered standard in the 31st century? Advertisements occur in dreams, everybody feels the way Fry felt outside of the Discount Shoe Outlet -- "I just saw something incredibly cool: A big, floating ball that lit up with every color in the rainbow, plus some new ones that were so beautiful I fell to my knees and cried." -- and robots envision their own realities even when they're not "jacking on."
Frankly, I'm impressed that Fry has adapted so well to such a weird future without chemical dependence. But then, he does have the support of a whole lot of friends. And Zoidberg.

OK. Now you've got me sparked up. *Ahem.* There are actual intoxicants, and then there are phony aphrodisiacs. Also, his name is Lrrr.
Rhino horn is commonly thought, in some current Earth cultures, to be an aphrodisiac. Rhinos are therefore poached nearly to extinction. I have always assumed that is what the alien poachers are doing to humans. (Though why "PROBE 1" is a useful vanity plate escapes me. Heh. Heh-heh. OK, I get it.)
Also, I have frequently wondered, as did Bender and Lrrr, why the nose horn (of the rhino or of Fry) was supposed to be the aphrodisiac, rather than the lower horn (of the rhino or of Fry.) But _Futurama_ and Lrrr (and Bigfoot) have convinced me that poaching is wrong, and that Ndnd can be pleased in other ways.
And as a guy with a big lower horn (what have you heard?), I'm very glad about that.
